Technique and Procedure

    Focusing with the High-Dry Objective

With your specimen in focus under low power and with your microscope properly Kohlered, you can now move to the "high-dry" (40X) objective. Just move the objective into place. Do not adjust the focusing controls before doing this. It is good technique to always observe the stage from the side to make sure that the objective will clear the slide. The objective lens should never touch the slide! Since your microscope is parfocal, meaning once the specimen is focused under one objective, it will be in aproximate focus under other objectives, the specimen should still be visible and still be in approximate focus. Adjust the fine focus a bit to bring the specimen into sharp focus.

To bring the specimen into sharp focus, adjust the fine focus knob a bit.
